In an Instagram post, the King and Queen wrote: "Happy birthday to the Prince of Wales!"
It featured as a caption of a new image showing a relaxed William sitting in a field.
The monarch’s message led message from other members of the Royal Family.
Moments later, Princess Kate and her children, Prince George, Princess Charlotte and Prince Louis have shared their own message.
Their post showed Prince William lay alongside their family dog Orla's never before seen adorable new puppies.
The image was taken by Kate in Windsor earlier this month.

They family captioned the post: “Happy birthday! Love C, G, C, L, Orla and the puppies!"
The message was signed off with a pair of paws emoji.
Orla is the Wales family’s black Cocker Spaniel and was a gift Kate's brother, James Middleton, in 2020.
William was born on 21 June 1982 at St Mary's Hospital in Paddington.

The Prince has been busy with royal duties in the run up to his birthday, taking a central role at several major events.
William was a key presence in last weekend's Trooping the Colour ceremony as well at the Order of the Garter ceremony and Royal Ascot earlier this week.
During Trooping the Colour, William took a prominent position alongside Princess Anne and the Duke of Edinburgh, all three riding on horseback for the monarch's official birthday parade.
The Prince of Wales wore military attire, including Full Ceremonial Guard Order of the Welsh Guards and traditional bearskin hat.

His resplendent uniform featured the Most Noble Order of the Garter Sash with Lesser George and a Welsh Guards Sword.
The Prince wore also wore gleaming medals on his chest, including the Gold, Diamond and Platinum Jubilee medals alongside the Coronation Medal.
While William rode in the parade down the Royal Mall, Princess Kate and their children rode by carriage.
At Royal Ascot on Wednesday, the Prince of Wales was present without Kate, who seemed to withdraw from the event at the last minute.

The Princess was said to be"disappointed" at missing the renowned races where William presented prizes on the second day of the festivities.
Kate's seemingly last-minute absence was soon revealed to have resulted from an simple admin blunder.
"Unfortunately, an inaccurate version of the carriage list was issued in error after that notification. The correct list was then circulated," a source told the Mail.
The administrative mix-up occurred when Ascot officials released the day's procession list at midday, listing both the Prince and Princess of Wales in the second carriage behind the King and Queen.
