The car Jota was travelling in is said to have derailed from the motorway, causing the fatal crash at around 00:30 local time on Thursday.
The Guardia Civil told BBC Sport that their Lamborghini left the road due to a tyre blowout while overtaking another vehicle on the road.
The car then reportedly caught fire.
The accident occurred during what should have been a celebratory period for the footballer, who had recently tied the knot with long-term partner Rute Cardoso.
The pair share three children together, making the tragedy even more devastating for the family.
